Etude des mécanismes d'action d'une immunothérapie par un lipide A, seul ou associé à l'oxaliplatine, dans des modèles de cancers coliques
2013
Colorectal cancer is a major public health concern in France. Resistance to standard chemotherapy requires development of novel therapeutic approaches. In the past decades, our team showed the immunotherapeutic properties of lipid A in a model of colon cancer in rats. 95% of rats bearing small carcinomas were cured following treatment by lipid A. The study of mechanisms underlying this immunotherapy allowed us to show that the antitumor effect of lipid A was dependent on cytotoxicity induced by granzyme B produced by intratumoral neutrophils. Indeed, we have shown that, in the tumor microenvironment, neutrophils produced granzyme B and had a pro-tumorigenic N2 phenotype. When rats were trea…
Caspase-1 involvement in colorectal cancer cell death phenomenons
2014
Colon cancer is a major public health matter because it has reached the second rank of cancer mortality in 2013 in France. Surgery, radiotherapy and standard chemotherapies helped by targeted therapies are not often efficient enough, underlying the need of using new therapies.Against this background, we have studied the possibilities to use new therapies (Liver X Receptor (LXR) ligands) or to improve classical treatment conditions (hyperthermic intraperitoneal chemotherapy (HIPC)) and the implication of caspase-1.We have first demonstrated that the nuclear receptor (LXR) agonists can induce pyroptotic cell death of colon cancer cell lines by inflammasome activation and subsequent caspase-1 …
Rôles des cellules myéloïdes suppressives et des infiltrats immunitaires dans le cancer
2013
Immune system plays a dual role in cancer: it can not only suppress tumor growth by destroying cancer cells, but also promote tumor progression by selecting immunoresistant tumor cells or by establishing an immunosuppressive microenvironment. Our main objective is to foster on the context of immune response in colon cancer settingDuring my thesis, I focused on one population of the immunosuppressive cells immune system: MDSC (myeloid derived suppressor cells). In this work, we explored strategies to reduce the number of these cells during tumor growth. We have discovered that small doses of 5 Fluorouracil are able to specifically induce apoptotic death of MDSC. We have characterized a novel…
